摘要
目的研究大鼠酒精性肝病模型的肾脏病理改变以及层粘蛋白,Ⅲ型胶原在肾脏的表达.方法以酒精灌胃法建立大鼠酒精性肝肾损伤的动物模型;应用 HE 染色,透射电镜观察其病理改变.免疫组化法检测16只对照组30只模型组的肾组织层粘蛋白及Ⅲ型胶原的表达.结果 HE 染色可见模型组肝脏出现酒精性肝病阶段性的病理改变.肾脏在相应阶段出现肾小球系膜增殖,问质局灶性炎细胞浸润,肾小管上皮细胞空泡变性或肿胀,管腔闭塞等改变.免疫组化染色,与同期对照组相比,酒精灌胃4wk、8wk,层粘蛋白表达无明显增加,差异无显著性,分别为 F_1=2.14,F_2=2.16(P>0.05).酒精灌胃12wk,层粘蛋白表达与同期对照组相比明显增加,F_3=3.68有显著性差异(P<0.05).各模型组与对照组相比Ⅲ型胶原表达无改变,两者无显著性差异F_1=2.08,F_2=2.11,F_3=2.10(P>0.05)结论长期持续的乙醇刺激,不仅可造成肝脏损害,又可引起明显的肾脏改变且以肾小管-间质损害为重.层粘蛋白在酒精性肾损害中表达增加,而Ⅲ型胶原无改变.
AIM To investigate the renal pathologic lesion of alcoholic liver disease in rats and the expression of laminin and collagen Ⅲ in kidney. METHODS The rat model of alcoholic hepatic and renal lesion by intragastric parfuaion of ethanol,then renal pathologic changes stained by hematoxylin and eosin stain were observed by electron microscope.The expression of laminin in 16 control rats and 30 model rats were evaluated by immunohistochemistry. RESULTS There were phasic pathologic changes in alcoholic liver disease in model rats.In the corresponding phase,the renal lesions presented glomerular mesangium proliferation,focal Inflammatory cell infiltration in tubulointeretitial,vacuolation or swell of renal tubule epithelium,obstruction of tubule.The expression of laminin in model rats are higher than that of control rats. There was a significant difference between the control and model rats(at the end of 12wk).F=3.68(P<0.05). There was no significant difference in the expression of collagen Ⅲ between the control and model rats.F_1=2.08, F_2=2.11,F_3=2.10(P>0.05). CONCLUSIONS Both hepatic lesions and significant renal pathologic changes could he induced by stimulation of ethanol for a long time.And changes of tubulointerstitial were more serious than that of glomerular.The expression of laminin in model rats were higher than that in control rats.There was no change in the expression of collagen Ⅲ in kidney.
