摘要
利用随机扩增多态性DNA (RAPD)在一个葡萄的种间杂交组合〔毛葡萄 83 4 96(Vi tisquinquangularis)×欧洲葡萄粉红玫瑰 (V .vinifera)〕的F1群体中发展分子标记 ,共产生了 89个稳定的RAPD标记 ,连同 4个形态标记 (花型、霜霉病抗性、果皮颜色、果汁颜色 )构建了一个葡萄RAPD分子连锁图。该图覆盖基因组总长度为 10 33cM ,标记间平均距离为 17.8cM ,为毛葡萄连锁图谱的构建提供了一个连锁框架。
A set of 280 random oligonucleotide perimers was screened for random amplified polymorphic DNAs(RAPD)fragments within a sample of 60 F 1 hybrids to generate RAPD markers and construct the molecular linkage map from two parents 83 4 96( V.quinquangularis )and Muscat Rose( V.vinifera ).89 repeatable RAPD markers as well as four morphological markers(flower type,grape downy mildew resistance,berry color and juice color)were obtained.The map consisted of 11 linkage groups,it covered a total genetic distance of over 1033cM(centimorgan),the average distance between markers was 17.8cM.It supplied a linkage framework for a more saturated linkage map of grape.
