小麦株高近等基因系的RAPD标记研究 被引量:9

摘要 采用 RAPD技术 ,以 4个小麦株高近等基因系 (分别含有 rht、Rht1、Rht2、Rht3基因 )为材料 ,筛选了 2 96个单一随机引物 ( 10个核苷酸 )。发现 2 5个引物的扩增产物在近等基因系间表现出特异性。在6次重复试验中 ,有 18个引物的特异扩增片段不能重复 ,6个引物可以重复 2~ 3次 ,唯有 OPAM0 1在全部试验中均能稳定重复 ,其特异扩增片段 OPAM0 11860 可以作为 rht基因的 RAPD标记。 Four near isogenic lines which contained rht, Rht1, Rht2, Rht3 respectively were used for polymorphic analysis at molecular level by RAPD technique. Out of 296 10 mer oligonucleotide primers screened, 25 showed polymorphism among near isogenic lines. During 6 repeat tests of these primers, 18 could not give the repeat results, 6 could repeat 2 to 3 times, only primer OPAM01 showed the stable results in which a 1.86 kb additional band was found in rht line. OPAM01 1860 could be identified as a RAPD marker linked to rht gene in wheat.
