微波辐射对孕鼠脾淋巴细胞活性氧和线粒体膜电位影响的研究 被引量:2

Study on the effect of microwave radiation on reactive oxygen species and mitochondrion membrane potential of splenic lymphocytes in pregnant rats

摘要 目的:研究微波辐射对孕鼠脾淋巴细胞中活性氧(ROS)活性和线粒体膜电位(△Ψm)的影响。方法:32只成年雌性Wistar孕鼠随机分为4组,每组8只,其中1组为对照组,3组为实验组;实验组分别采用辐射强度为2.5、5、10 mW/cm2,辐射时间6 min/次,5次/每周,连续辐射4周,对照组不辐射;采用流式细胞术,间接检测细胞内ROS活性和△Ψm。结果:辐射强度为2.5 mW/cm2组细胞内ROS活性和△Ψm与对照组差异无统计学意义(P>0.05);辐射强度5、10mW/cm2组的ROS活性较对照组增加,△Ψm较对照组降低,差异有统计学意义(P<0.05),且呈剂量-反应关系。结论:长期低剂量微波辐射能够诱导孕鼠脾淋巴细胞中ROS活性增加,而△Ψm降低,并且具有一定的剂量效应规律性。 Objective: To research the effect of microwave radiation on reactive oxygen species (ROS) activity and mitochondrion membrane potential ( △ψm) of splenic lymphocytes in pregnant rats. Methods: Thirty - two adult male Wistar pregnant rats were randomly divided into one control group and three experimental groups, eight rats in each group; the rats in experimental groups were radiated by mi- crowave respectively (2.5, 5, and 10 mW/cm2 ), the radiation time was six minutes per time, five times one week for continuous four weeks; the rats in control group didnt received radiation; flow cytometry was used to detect ROS activity and △ψm in splenic lymphocytes indirectly. Results: There was no statistically significant difference in ROS activity and△ψm between 2.5 mW/cm2 experimental group and control group ( P 〉 0. 05 ) ; ROS activities in 5, 10 mW/cm2 experimental groups were higher than that in control group, but △ψm lev- els were lower than that in control group, there were statistically significant differences ( P 〈 0. 05 ), showing a dose - dependent manner. Conclusion: Long - term low - dose microwave radiation can induce increase of ROS activity and decrease of △ψm in splenic lymphocytes of pregnant rats, showing a dose - dependent manner.
