摘要
目的探讨ADAM12、IGF-Ⅱ联合检测与自然流产的关系。方法放免法、时间分辨荧光法分别检测正常妊娠、先兆流产、稽留流产组血清IGF-Ⅱ、ADAM12-S水平;原位杂交法检测绒毛组织ADAM12-S、IGF-ⅡmRNA水平。结果①血清ADAM12-S、IGF-Ⅱ表达在正常妊娠、先兆流产、稽留流产依次下降,两两有显著差别(P<0.05)。②绒毛ADAM12-S、IGF-Ⅱ的mRNA表达在正常妊娠组、先兆流产组、稽留流产组依次下降,两两比较有显著性差别(P<0.05)。③血清ADAM12-S与IGF-II表达正相关(r=0.309),差异有统计学意义(P<0.05)。结论联合检测ADAM12和IGF-Ⅱ可提高预测自然流产的准确性,两者相互作用失衡可能是导致自然流产发生的重要原因。
Objective To study the relationship between combined test of ADAM12 and IGF- Ⅱ and spontaneous abortion. Methods The expression of Serum IGF- Ⅱ were measured by radio immunoassay using commercial kit in normal early pregnant women,threatened abortion women and missed abortion women. The expression of serum ADAM12-S were measured by Time- resolved detection technology. Immunohistochemistry and in-situ hybridzation were used to investigate the expression of IGF- Ⅱ mRNA and ADAM12-S mRNA in villus. Results (1) Order of suc- cessively decreasing expression of serum ADAM12-S and IGF- Ⅱin normal early pregnant women, threatened abor- tion women and missed abortion women. There were significant differences between the two two (P 〈 0.05).(2) Order of successively decreasing expression of villus ADAM12-S and IGF- Ⅱ mRNA in normal early pregnant women, threat- ened abortion womenand missed abortion women. There were significant differences between the two groups(P 〈 0.05 ). (3)ADAM12-S and IGF-Ⅱ in maternal serum expression levels were positively correlated, r = 0.309, with statistical significance (P 〈 0.05). Conclusion The combined detection of ADAM12 and IGF- Ⅱ can improve the accuracy of spontaneous abortion, the interaction between imbalance may be important causes of spontaneous abortion.
