摘要
[目的]研究新西兰白兔肾性高血压模型眼底并发症的指标变化。[方法]利用"双肾一夹"方法制备新西兰白兔高血压模型。术后每周监测血压并观察眼底。5个月后检测视网膜组织一氧化氮(Nitric oxide,NO)、丙二醛(Malondialdehyde,MDA)、超氧化物歧化酶(Superoxide dismutase,SOD)、一氧化氮合酶(Nitric oxide synthase,NOS)、内皮素(Endothelin 1,ET-1)、谷氨酸(Glutamate,Glu)、玻璃体腔表皮生长因子(Epidermalgrowth factor of vitreous,EGF)、基质金属蛋白酶-9(Matrix metalloproteinase9,MMP-9)、血管性血友病因子(von Willebrand factor,vWF)及细胞间粘附分子-1(IntercellularAdhesion Molecule-1,ICAM-1)含量。左眼球病理切片。[结果]术后4个月模型组血压升高;眼底出现视网膜棉绒状斑,小动脉变细,动脉毛细血管稀少等现象;视网膜NO、SOD、Glu含量较低;而MDA、iNOS含量较多,其它指标无明显差异;视网膜内核层、外核层细胞排列疏松空隙较大,神经节细胞较少,外界膜组织排列紧密染色较深,巩膜组织呈列无序较松散。[结论]高血压模型兔眼底视网膜、小动脉发生病变,可能与组织抗氧化、血管舒张能力降低,组织毒性物质含量增加及兴奋性氨基酸Glu含量降低有关。提示"双肾一夹"型新西兰白兔肾性高血压模型可造成眼底缺血缺氧,进而引起一系列眼底病变。
[Objective] To study fundus complications due to hypertension in rabbits by observing the changes of vessels ,biochemical indexes, parrs Cytokine and Organization Form of retina.[Methods] We made rabbits renal arterial stenosis by surgery to make hypertension model.Indexes:Measured blood pressure and fundus observation once a week after surgery .5 months after surgery, extirpated the left eyes and peeled retina after anaesthesia, and the activities of nitric oxide(NO), malondialdehy(MDA), superoxide dismutase(SOD), nitric oxide synthase(NOS) , Endothelin I(ET-1), Glutamate(Glu), Epidermal Growth Factor of vitreous(EGF), matrix metanoproteinase9(MMP-9), von Willebrand factor(vWF), Intercellular Adhesion Molecule-1 (ICAM-1) were measured with pathologic examination. [Results]The blood pressure of model rabbits was higher than that of normal rabbits 4 months after surgery, and some lint spots appeared on retina of model rabbits, most arterioles tended to be thin and the number of arterial capillary became less. Compared to normal rabbits,in retina homogenate of hypertension model rabbits,there were less NO,SOD and Glu, oppositely, there were more MDA and iNOS ,however the other indexes had no difference between normal and model rabbits. In retinal pathological sections of hypertensive rabbits, the structure became changed. [Conclusion] The retina and arteriosclerosis of hypertensive rabbits would be un-normal .It maybe relate to the retinal damage including the lower of antioxidant and vasodilation capacity, and the higher level of cytotoxic substance like MDA and iNOS , in addition the shortage of Glu may also relate to retinal damage. Relying on the result we can predict that the fundus complications may be caused by hypoxic ischemia made by hypertension.
