大黄素和大黄多糖对铜负荷模型大鼠脂质过氧化的影响 被引量:5

Effect of Emodin and Rheum Tanguticum Polysaccharide on Lipid Peroxidation in Copper-Overloaded Rats

摘要 目的探讨大黄素(EMD)和大黄多糖(RTP)对铜负荷模型鼠体内脂质过氧化以及肝功能的影响。方法将60只Wistar大鼠随机分为对照组、模型组和给药组,每组20只,以铜负荷饮食法造模8周,造模第4周后给药组给予EMD、RTP各200mg/(kg·d)灌胃,对照组和模型组给予等量生理盐水,各组共灌胃4周。观察各组大鼠血清和肝组织丙二醛(MDA)、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)水平和肝功能指标的变化。结果与对照组比较,模型组血清和肝组织中MDA水平显著升高,SOD、GSH-Px水平显著下降(P<0.05或P<0.01);模型组血清丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)和总胆红素(TBiL)水平较对照组显著升高,白蛋白(ALB)水平显著下降(P<0.01)。给药组较模型组血清和肝组织中MDA水平显著下降,SOD、GSH-Px水平显著升高(P<0.05或P<0.01);给药组ALT、AST和TBiL水平较模型组显著下降(P<0.05或P<0.01)、ALB水平无显著变化(P>0.05)。结论 EMD联合RTP能抑制铜负荷模型鼠体内脂质过氧化,改善其肝功能水平。 Objective To research the effect of Emodin (EMD) and Rheum tanguticum polysaccharide (RTP) on lipid peroxidation and liver function in copper-overloaded rats. Methods Sixty Wistar rats were randomized into the control group, model group and treatment group, with 20 in each. The rat models were established with copper-overloaded diet for eight weeks. The treatment group was intragastrically administrated with 200 rag/ (kg· d) EMD and RTP. The other two groups were given normal saline. The treatment lasted for four weeks. The levels of malondialdehyde (MDA), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), alanine aminotransferase (ALT), aspartate aminotransferase (AST), total biliru- bin (TBiL) and albumin (ALB) Were observed. Results Comparing with the control group, the levels of MDA, ALT, AST and TBiL were significantly increased (P〈0.01) but the levels of SOD, GSH-Px and ALB were significantly decreased (P〈 0.05 or P〈0.01) in the model group. Comparing with the model group, the levels of SOD and GSH-Px were significantly increased (P〈0.05 or P〈0.01) but the levels of MDA, ALT, AST and TBiL were significantly decreased (P〈0. 05 or P〈 0.01) in the treatment group. There was no significant difference between the model group and treatment group in the ALB level (P〈0.05). Conclusion The combination of EMD and RTP can inhibit the lipid peroxidation and improve liver function in copper-overloaded rats.
