摘要
目的 :运用小剂量四氯化碳 (CCl4)豆油溶液皮下注射结合高脂饮食制作大鼠非酒精性脂肪肝 (NAFL)模型 ,找出既能成功制造大鼠NAFL模型而又对大鼠肝功能影响较小的造模方法。方法 :用CCl4豆油溶液皮下注射结合高脂饮食造模 ,按体重计前 3周给予 40 %CCl4豆油溶液 0 2ml/10 0 g ,后 3周给予 40 %CCl4豆油溶液 0 1ml/10 0g ,同时与常用剂量 40 %CCl4豆油溶液 0 3ml/10 0g皮下注射结合高脂饮食所造模型进行肝功能、血脂和肝脏脂质积沉等方面的比较。结果 :6周后 ,小剂量CCl4豆油溶液皮下注射结合高脂饮食造模不仅能使血TG、TC和肝脏TG含量显著升高 ,且与常用剂量CCl4豆油溶液皮下注射结合高脂饮食所造模型相比 ,肝功能损伤更小。结论 :持续 6周的小剂量CCl4豆油溶液皮下注射结合高脂饮食喂养既能制造出比较满意的NAFL动物模型 ,又能显著减少造模时的肝功能损害 ,是相关研究造模的一种可行性选择。
Objective: To analyze the rat models of nonalcoholic fatty liver (NAFL) which were made by a compound method of injecting CCl 4 hypodermically in smaller dose twice a week and taking food full of fat daily at the same time, and to investigate the effects of the method on the alteration of the liver of those rat models. Methods: The smaller dose group of rat NAFL model was made by a compound method of injecting 40% CCl 4 hypodermically twice a week in a 0.2ml/100g dose in the previous 3 weeks and in a 0.1ml/100g dose in the later 3 weeks, and the other group by injecting 40% CCl 4 hypodermically twice a week in a 0.3ml/100g dose, and at the same time taking food full of fat daily for 6 weeks. Then, 6 weeks later, all rats were killed. And liver function, the content of serum TC, TG and HDL-C, and the content of TG and TC in liver tissues were measured. Then those data were analyzed by software SAS 8.1(TS1M0) using F test and q test. Results: The smaller dose group could not only promote the content of TG in liver tissues and the content of serum TG and TC 6 weeks later, but also make less lesion with liver function than that method which were injecting CCl 4 hypodermically in a common dose such as 0.3ml/100g twice a week and taking food full of fat daily. Conclusion: Injecting CCl 4 hypodermically in smaller dose twice a week and taking food full of fat daily at the same time for 6 weeks can not only make satisfied rat models of NAFL but also diminish the lesion of liver function. So we deem that it is a good choose for correlative studies.
