摘要
目的:探讨不同浓度苦参水提液对细胞增殖的影响。方法:采用四甲基偶氮唑蓝(MTT)比色法检测不同浓度(5、2、1、0.5、0.05、0.025、0.005 mg/mL)的苦参水提物在不同的时间点(24、48、72 h)对HepG-2、L02细胞增殖的影响。10 g/(kg.d)的苦参水提液予ICR小鼠灌胃,10%含药血清培养L02细胞,MTT比色法检测不同的时间点(24、48、72 h)对细胞增殖的影响。以△OD值(各组加药平均值-正常组平均值)表征抑制或促进作用。结果:苦参水提物浓度≤2 mg.mL-1时对细胞增殖有一定促进作用,尤其以0.5、1 mg.mL-1的促进作用最明显(P<0.01),当苦参水提物浓度达到2 mg.mL-1,处理72 h时,细胞增殖出现抑制作用。含药血清能显著促进L02细胞的增殖。结论:苦参水提物在一定浓度范围内促进细胞增殖,高浓度抑制细胞增殖。
Objective ： To explore the promotion effects of water extracts from Sophora flavescens on proliferation of hu- man embryonic liver cell L02 and liver cancer cell HepG2 cells. Method：MTF assay was used to determine the effects of different time （24,48 ,72 h）incubation on the proliferation of on HepG -2 and L02 cells with different concentrations （5.0,2.0,1.0,0.5,0.05,0.025,0. 005 mg· mL^- 1 ） of water extracts from Sophora flavescens. 10 g/（ kg · d） of the water extracts of Sophora flavescens. Mice was fed ICR to make the drug serum. L02 cells were cultured with 10% drug serum. The effects of the serum on L02 cells proliferation was detected by MTF assay at 24 h and 72 h. The data was an- alyzed by T - test. Result ： MTT analysis indicated that proliferation of the L02 and HepG2 cells were promoted markedlyby the aqueous extracts from Sophora flavescens in a dose - dependent manner when the drug concentration 〈 2.0 mg · mL^- 1. The promoting effects was significant when the drug concentration at 0.5 （ P 〈 0.05 ） and 1.0 mg · mL^- 1 （ P 〈 0.01 ）. The inhibition effects of the extracts on the two kinds of cells was shown when the cells was treated with the ex- tracts at 2.0 mg · mL^- 1for 72 h. 5 mg · mL^- 1 aqueous extracts significantly inhibited the cells proliferation. L02 cells prolif- eration was significantly promoted by the serum of mice administered the aqueous extracts from Sophora flavescens. Con- clusion ： The low concentration of the aqueous extracts from Sophora flavescens promoted the cells proliferation but the high concentration of the aqueous extracts inhibited the cells proliferation.
