摘要
目的:进一步探讨复方苦参含药血清对人结肠癌细胞SW480的凋亡诱导作用。方法:采用血清药理学方法研究复方苦参对结肠癌细胞SW480的体外效应。观察含药血清对SW480细胞克隆形成率的影响,应用荧光染色,流式细胞仪及透射电镜观察含药血清对SW480的凋亡诱导作用。结果:含药血清对SW480细胞克隆形成率有一定抑制作用。含药血清作用细胞48h后,可观察到胞核固缩、荧光染色增强、胞核碎裂等凋亡形态学变化,流式细胞仪检测到亚G1峰。结论:复方苦参含药血清体外抑制SW480细胞的增殖并能诱导其凋亡,诱导凋亡是其抗肿瘤的机制之一。
Obuective To explore the effect of Radix Sophorae Flavescentis （KCP） contained serum in inducing apoptosis of human colon cell line SW480 Methods Methods of sero - pharmacology were taken in the study. The effect of KCP contained serum on cloning efficiency in SW480 were evaluated. Apoptosis of SW480 were observed by fluo- rescent staining, transmission electronic microscopy and flow cytometry（FCM）. Results KCP contained serum could inhibit the cloning efficiency of SW480. After treated by KCP contained serum for 48 hours, the cells showed features of apoptosis and Sub- GI peak was detected by FCM. Conclusion In vitro, KCP contained serum can inhibit cell proliferation and induce apoptosis of SW480. Inducing apoptosis maybe one of the anti-neoplasm mechanism of the medicine.
