鹿茸饮片的DNA条形码鉴别研究 被引量:44

摘要 目的建立一种快速、准确和标准化的鹿茸饮片DNA条形码鉴别方法,并分析鹿茸药材原动物间的亲缘关系。方法通过下载GenBank鹿类动物线粒体细胞色素C氧化酶亚基Ⅰ(COⅠ)基因,经比对分析,在靠近COⅠ基因5′端设计了一对鹿科动物DNA条形码通用引物,对来源于鹿科动物3个属共计19份样本的鹿茸进行了PCR扩增和序列分析,并构建系统进化树。结果通过遗传距离和分子系统树分析显示DNA条形码鉴别方法能够在属水平和种水平将鹿科3属9种鹿成功的鉴别开来,对90%的鹿茸样本可进行有效地鉴定,而梅花鹿、马鹿中各一个样本未得到准确鉴定,具体原因有待进一步研究。结论所建立的鹿茸药材DNA条形码鉴别方法,准确可靠,可用于正品鹿茸马鹿和梅花鹿及其混伪品进行准确鉴定。 OBJECTIVE To establish a rapid, accurate and standardized method for identifying Cornu Cervi Pantotrichum with DNA barcoding method and analyze the genetic relationship of origin animals of Cervidae. METHODS A pair of universal primer was designed according to the sequence alignment of COI genes. And then it was carried out the PCR amplification reaction, sequence analysis for 19 samples from cervi animals. RESULTS The establishment of DNA barcoding technique could successfully identify 9 species out of 3 genera of Cervidae at specie level by sequence alignments, genetic distance analysis and phylogenetic tree analysis. Resultss showed that 90% samples of Cornu Cervi Pantotrichum could be effectively identified except few samples of Cervus Nippon and Cervus elaphus. CONCLUSION DNA barcoding identification method is accurate and reliable and could be applied to identify the genuine and false samples of Cornu Cervi Pantotrichum in markets.
