摘要
采用响应曲面分析法对超声波提取灵芝多糖的工艺条件进行优化,优化后的工艺为:超声功率320W,提取温度70℃,提取时间34 min,其多糖得率为2.78%,氧自由基吸收能力(ORAC值)为1956μmolTrolox/g,与理论预测值无显著性差异。因此,采用响应面分析法优化灵芝多糖超声提取工艺,具有较好的可行性。在最佳条件下,制备的灵芝多糖分子量组成为18150 Da(20.07%)和6484 Da(79.93%),大大低于传统水提灵芝多糖的分子量。
The response surface methodology was used to optimize the ullrasonic extraction of Ganoderma lucidum polysaccharides. Design expert analysis indicated that the optimal processing conditions were: ultmsounic power 320 W, temperature 70 ℃ and extraction time 34 rnin, respectively. The extraction yield and the ORAC value of polysaccharide were 2.78% and 1956 lamol Trolox/g, respectively, which had no significant differences with the predicted values, indicating that the response surface methodology was a reasonable method to optimize the extraction conditions of Ganoderma lucidum polysaccharides. The molecular weights dislribution of the polysacchaddes under the optimal conditions were 18150 Da (20.07%) and 6484 Da (79.93%), much less than those of the polysaccharides prepared by traditional water extraction.
