地黄饮子对海马神经元缺氧损伤的保护机制 被引量:13

摘要 目的观察地黄饮子(熟地黄、巴戟天、山茱萸、石斛、肉苁蓉等)对缺血缺氧损伤海马神经细胞的保护作用并探讨机制。方法将培养细胞分为对照组、模型组及药物保护组;其中,损伤组采用Na2S2O41 mmol/L加低糖作用3 h;药物保护组在加入损伤因素同时加入不同浓度含地黄饮子血清。通过比色法测定培养液中乳酸脱氢酶含有量(LDH),采用MTT法测定脑海马神经细胞存活率,利用线粒体内琥珀酸脱氢酶(SDH)含有量来分析地黄饮子的保护作用及其作用的有效浓度;测定缺血缺氧损伤下海马神经细胞内超氧化物歧化酶(SOD)与丙二醛(MDA)含有量。结果含地黄饮子血清与空白血清比例在1∶3~3∶1之间,均对缺氧神经细胞具有保护作用。降低损伤细胞LDH释放,细胞膜损伤程度减轻。MTT值不同程度升高,提示细胞活力状态提高、细胞能量代谢过程有所改善;地黄饮子可以保护缺血缺氧环境中海马神经细胞内SOD的活力,降低MDA的含有量,而且随剂量的增加而增加。结论地黄饮子对缺氧损伤脑海马神经细胞有明显保护作用;其作用机制可能与降低氧自由基损伤有关。 AIM To observe the protective effect and mechanism of Dihuang Drink (Rehmanniae Radix praeparata, Morindae officinalis Radix, Corni Fructus, Dendrobii Caalis, Cistanches Herba, etc. ) on the anoxaemia of cultured hippocampal neurons. METHODS Cultured hippocampal neurons were divided into three groups: control group, model group and drug-protective group. Hypoxia induced by 1 mol/L Na2S204 in low sugar solution on hippocampal neurons for 3 h in the model group, while different concentrations of Dihuang Drink were added at the same time in the drug-protective group. Colorimetric method was used to test the concentration of delectate hydroge- nase (LDH) , and MTF method was applied to observing the hippocampus cell survival. The content of succinic dehydrogenase (SDH) in mitochondria was intended to reflect the protection and effective concentration of Dihuang Drink. Moreover, the changes of malondialdehyde (MDA) and superoxide dismutase (SOD) were measured in the cultured hippocampal neurons. RESULTS Dihuang Drink had protective effect on hippocampal neurons at ratio of the concentration of Dihuang Drink to blank serum ( 1 : 3 to 3 : 1 ). Decrease in the release of LDH could reduce the damage to cell membrane. The increase in MTT value meaned cell activity was enhanced and cell energy metabolism was improved. Moreover, Dihuang Drink concentration-dependently increased the content of SOD and de- creased the MDA (P 〈 0. 01 ) in the hypoxic hippocampal neurons. CONCLUSION Dihuang Drink has an obvious protective effect on the hypoxic hippocampal neuron via decreasing free radicals injury.
