摘要
目的观察脑灵汤对阿尔茨海默病(Alzheimer'sDisease,AD)大鼠模型学习记忆能力及脑内海马细胞凋亡基因Bcl-2和Bax表达的影响。方法通过半乳糖腹腔注射及氯化铝(Alcl3)皮下注射建立AD大鼠模型,采用γ-电迷宫检测学习记忆能力,提取脑内海马组织总RNA,应用RT-PCR和琼脂糖凝胶电泳方法测定Bcl-2、Bax基因的表达。结果脑灵汤能改善模型大鼠的学习记忆能力,增强受损海马神经元Bcl-2基因的表达,降低Bax基因的表达。结论脑灵汤可能通过升高Bcl-2/Bax的比值而抑制海马神经元的凋亡,改善模型大鼠学习记忆能力。
[Objective] To investigate the effect of Nao ling decoction to the expression apoptosis related gene bel-2 and bax of the hippoeampus cells in rats with Alzheimer's disease and it's behavior of learning and memory. [Method] The AD model of rats was established by be given D-galaetose, ip;and Alel3, ig, Y-electric morris maze was used to test learning and memory; Distilled the total RNA and the expression of bcl-2 and bax by using the methods of Reverse Transeription-Polymerase Chain Reaction and agarose geleleetrophoresis were measured. [Resuit] The combined use of D-galaetose and Alcl3 induced some alterations in rats, such as learning and memory impairment, The Nao ling decoction can enhance the expression of bel-2 gene ,reduce the expression of bax gene in- jured hippoeampal neurons. [Conclusion] The Naoling decoction enehaneed the ratio of bel-2/bax, to restrain the apoptosis of the hippoeampal neurons, improve the leaning and memory of the AD' rats.
