摘要
目的研究全新丹参素衍生物ADTM对心肌细胞凋亡的影响。方法体外培养H9c2心肌细胞,t-BHP诱导H9c2细胞凋亡,Hoechst染色观察ADTM对心肌细胞凋亡的影响;结扎大鼠左冠状动脉前降支建立大鼠心肌缺血模型,缺血24h后提取心肌组织蛋白,Western blot方法检测心肌组织凋亡相关蛋白Bcl-2、Bax以及caspase-3的表达。结果 ADTM能显著抑制t-BHP诱导的H9c2心肌细胞凋亡;在大鼠心肌缺血模型中,明显提高Bcl-2蛋白的表达,降低Bax和caspase-3蛋白的表达,减少缺血心肌细胞凋亡。结论 ADTM可显著抑制心肌细胞凋亡,从而减轻心肌细胞缺血损伤。
Objective To investigate the in vitro and in vivo anti-apoptosis effect of ADTM, a novel derivative of Dansh- ensu, on cardiomyocytes. Methods Hoechst 33258 staining was used to detect H9c2 cell apoptosis induced by t BHP in vitro experiment. A rat model of acute myocardial ischemia was established by the ligation of left coronary artery an- terior descending branch, and the expression of apoptosis-related protein Bcl-2, Bax and caspase-3 was measured by Western blot. Results ADTM significantly inhibited the H9c2 cell apoptosis. ADTM up-regulated the expression of Bcl-2 protein and down-regulated the expression of Bax and caspase-3 in a dose-dependent manner. The biological activity of ADTM was remarkably better than Danshensu. Conclusion Inhibition of cardiomyocytes apoptosis may be a potential protective mechanism of ADTM against cardial ischemia injury.
