摘要
采用日本血吸虫重组BCG-Sj26GST疫苗免疫BALB/c小鼠。免疫8周时,断头取血,取脾脏,取腹腔巨噬细胞。以淋巴细胞刺激指数(SI)反映细胞增殖能力,以NO湫得检测巨噬细胞吞噬活性,以ELISA试剂盒检测血清及脾淋巴细胞培养上清的白细胞介素2(IL-2_和干扰素γ(IFN-γ)的含量。结果表明,rBCG-Sj26GST疫苗以10^-6CFU剂量经皮下免疫小鼠后。
In order to investigate the immunogenicity of recombinant BCG bearing Shistosoma japon- icum 26 ku glutathione S-transferase gene (rBCG -Sj26GST), the specific pathogen-free BALB/c mice were immunized with rBCG-Sj26GST vaccine in dose of 10~6 CFU by SC. The lymphocyte stimulating index (SI), macrophage activity as well as IL-2, IFN-γ levels of the serum and culture supernatant of spleen lymphocytes were tested in the killed mice on the 8th week of inoculation. The results showed that SI was significantly higher than those in the control, vector and BCG groups (P<0. 05). The macrophage activity was significantly higher than in the control and vector groups (P<0. 01). The serum IL-2 level was significantly higher than in the control group (P<0. 01) and vector group (P<0. 05). Compared with control, vector and BCG groups, the IL-2 level of the culture supernatant of spleen lymphocytes was increased by 44%, 48% and 42% respectively, and the serum IFN-γ level by 20%, 19% and 13% respectively. These findings suggest that the rBCG-Sj26GST vaccine could obviously enhance the immune response of the BALB/c mice, which is related with the immun adjuvant activity of BCG, on the other hand with specific anti-Sj26GST antibody from BALB/c mice.
