烧伤大鼠淋巴液细胞因子及淋巴结T淋巴细胞亚群的变化 被引量:4

Changes in cytokines in lymph and T lymphocyte subsets in lymph node of burned rats

摘要 目的 观察烧伤对大鼠淋巴液细胞凶子及淋巴结T淋巴细胞亚群的影响。 方法取18只Wistar大鼠,将每只鼠单侧后腿浸于70℃热水30 s,造成约4% TBSA深Ⅱ度烫伤(烧伤组);对侧后腿浸于22℃温水30 s模拟烫伤(假伤组)。分别于伤后6、24、72 h按随机数字表法取6只大鼠,收集2组髂总淋巴结及其输出淋巴管内的淋巴液,ELISA法检测淋巴液中TNF-α、吖干扰素、IL-4水平并计算叫干扰素111-4比值,流式细胞术检测淋巴结中CD4+、CD8 +T淋巴细胞百分比并计算CD4 +/CD8+比值。对数据进行z检验。 结果 (1)大鼠伤后6、24、72 h,烧伤组淋巴液TNF-α水平分别为(51.6±5.4)、(27.4±2.6)、(23.O±2.7)pg/mL,明显高于假伤组的(17.8±1.6)、(16.4±1.2)、(17.2 +2.0) pg/mL,t值分别为15. 346、11. 854、4.189,P值均小于0.叭。(2)伤后6、24、72 h,烧伤组与假伤组吖干扰素水平比较,差异无统计学意义(t值分别为2. 059、-0. 805、-0. 415,P值均大于0. 05);烧伤组IL-4水平明显高于假伤缉(t值分别为9.141、11. 669、6.940,P值均小于0.01),吖干扰素/IL-4比值(2.27 +0. 34、1.54±0.19、1.60±0.16)明显低于假伤组(3. 33±0.25、3.34±0.22、2. 52±0.24,t值分别为-6. 298、- 11. 313、-8. 893,P值均小于0.01)。(3)伤后6、24 h,烧伤组与假伤组CD4+、CD8 +T淋巴细胞百分比及CD4 +/CD8+比值比较,差异无统计学意义(t值为-2. 48-0. 215,P值均大于0.05)。伤后72 h,烧伤组CD4 +T淋巴细胞百分比及CD4 +/CD8+比值分别为(38.6±2.3)%、2. 13±0.16,明显低于假伤组的(48.9±2.9)%、2. 68±0.12,t值分别为-7.551、-5. 068,P值均小于0.01;CD8 +T淋巴细胞百分比与假伤组比较,差异无统计学意义(t=0.845,P〉0. 05)。 结论 烧伤能够引起大鼠局部引流淋巴液吖干扰素/IL-4比值和局部引流淋巴结内CD4 +/CD8+比值降低,抑制其局部免疫功能。 Objective To observe the effect of burn on c:ytokines in lymph ancl T lymphocyte suh- sets in lymph node of rats. Methods Eighteen Wistar rats were used in the experiment. One of the hind limbs of each rat was immersed in 70 cC hot water for 30 s to reproduce 4% TBSA deep partial-thickness scald model ( burn group) , while the other hind limb was immersed in 22 cC warm water for 30 s to simulate scalcl ( sham injury group). On post injury hour ( PIH) 6, 24, and 72, 6 rats were chosen accorcling co che random numher table. Lymph fluid in the lymph vessel of each animal ( two groups) was obtained for decer- mination of levels of tumor necrosis factor a ( TNF-a) , interferon-y ( IFN-y) and interleukin-4 ( IL-4) by ELISA, and IFN-y/IL-4 ratio was calc:ulated. Common iliac lymph node of eac:h animal ( two groups) was obtained for determination of ratios of CD4 + , CD8 + T lymphocytes with flow cytometry, and CD4 +/CD8 + ratio was calculated. Data were proc:essed with t test. Results (1) On PIH 6, 24, and 72, TNF-a level in burn group was respectively ( 51. 6 + 5. 4 ) , ( 27.4 + 2. 6) , ( 23. 0 + 2. 7 ) pg/mL, which were signifi- candy higher than those in sham injury group [ ( 17. 8 + 1. 6) , ( 16. 4 +1.2) , ( 17. 2 +2. 0) pg/mL, with t value respectively 15. 346, 11. 854, 4. 189, P values all below 0.011. (2) On PIH 6, 24 , and 72, Lhere was no significant statistic:al difference between burn group and sham injury group in IFN-y level ( with t val- ue respectively 2. 059 , - 0. 805 , - 0. 415 , P values all above 0. 05 ) ; IL-4 level in burn group was respec-tively higher than that in sham injury group ( with t value respectively 9. 141 , 11. 669 , 6. 940, P values all below 0. 01) ; IFN-y/IL-4 ratio in burn group ( 2. 27 + 0. 34 , 1. 54 + 0. 19 , 1. 60 + 0. 16 ) was respectively lower chan thac in sham injury group (3. 33 +0. 25 , 3. 34 + 0. 22, 2. 52 + 0. 24, with t value respectively -6. 298, - 11. 313, -8. 893, P values all below 0. 01). (3) On PIH 6 and 24, chere was no significant statiscic,al clifference between burn group ancl sham injury group in ratios of CD4 + and CD8 + T lymphocytes anci also CD4 +/CD8 + ratio ( with t values from - 2. 486 to - 0. 215 , P values all above 0. 05 ) . On PIH 72 , ratio of CD4 + T lymphocytes and CD4 +/CD8 + ratio in burn group was respectively (38. 6 + 2. 3)% and 2. 13 +0. 16, which were significantly lower than those in sham injury group [ (48.9 +2. 9) % and 2. 68 + 0. 12, with t value respectively - 7.551, - 5.068, P values below 0.01 ] ; there was no significant statisti- cal difference between burn group and sham injury group in ratio of CD8 +T lymphocytes ( / = 0. 845 , P 〉 0. 05) . Conclusions Burn may decrease IFN-y/IL-4 raLio in loc:ally drained lymph and CD4 +/CD8 + ra- tio in locally drained lymph node of rat, which may inclicate lowering of local immune function.
