摘要
以"月月红"(R.chinensis‘Slater’s crimson China’)的嫩叶为外植体,采用MS为基本培养基,在添加不同浓度的2,4-D、TDZ、GA3和NAA上,结合不同时间的暗培养,研究"月月红"胚性愈伤组织的诱导及植株再生。结果表明,2,4-D 5 mg/L暗培养20 d诱导出的愈伤组织,在MS+TDZ 1.0 mg/L+GA30.1 mg/L+NAA0.01 mg/L培养基上暗培养12 d,可以获得较好的胚性愈伤组织;胚性愈伤组织转入光照培养15 d后可形成肉眼可辨的子叶期体胚及具有不定芽和胚根的结构,20 d后可长成绿色的小植株。
The effects of different concentrations of 2,4-D,TDZ,GA3,NAA,combination with the condition of illumination on the status of embryogenic callus induced from young leaves of R.chinensis 'Slater's crimson China'were studied and the embryogenesis and plant regeneration of embryogenic callus were also investigated.The result showed that callus could be induced by the media MS+2,4-D 5mg/L + proline 300 mg/L.The best media for embryogenic callus induction was MS+TDZ 1.0 mg/L+GA3 0.1 mg/L+NAA 0.01 mg/L for 12 dark days.Cotyledonary embryoid and tissues structure of adventitious buds and embryonic root were obtained from embryogenic callus cultured under light for 15 days,and planted young plants for 20 days.
