红金钻蔓绿绒高频再生体系的建立 被引量:7

The Establish of High Frequency and Regeneration System for Philodendron ‘Red Congo’

摘要 以红金钻根茎、叶片、叶柄为外植体,通过直接诱导不定芽和间接(愈伤组织)诱导不定芽途径,建立组培快繁体系。本研究发现:根茎直接诱导不定芽最佳培养基:MS+3 mg/L 6-BA+0.2 mg/L NAA+2 mg/L GA_3;通过愈伤组织间接诱导不定芽及其分化的最佳培养基:MS+1.0 mg/L 6-BA+0.2 mg/L IBA;不定芽增殖最佳培养基:MS+3.0 mg/L 6-BA+0.8 mg/L IBA;生根最佳培养基为:1/2MS+0.75 mg/L NAA。将植株种植于草炭和珍珠岩以3:1混合的基质中,成活率达98%。 By taking the rhizome,leaf and petiole of the Philodendron‘Red Congo’as explants,the tissue culture and rapid propagation system was established through the method of direct and indirect(callus tissue)to induce adventitious bud.The results showed that:the optimal medium for direct inducement on adventitious bud by rhizome was that MS+3 mg/L 6-BA+0.2 mg/L NAA+2 mg/L GA3.The optimal medium for indirect inducement on adventitious bud by callus and for the callus differentiation was that MS+1.0 mg/L 6-BA+0.2 mg/L IBA.The best medium for adventitious bud multiplication was that MS+3.0 mg/L 6-BA+0.8 mg/L IBA.Finally,the optimal medium for rooting was that 1/2MS+0.75 mg/L NAA.The survival rate of these plants could reach 98% by being planted in the substrate of turf and perlite at the rate of 3:1.
