摘要
针对常用乳糖测定方法存在的局限性和不便性,结合高效毛细管电泳(HPCE)法简单快速、高效低耗的特点,探索出了高效毛细管电泳仪配合紫外检测器测定乳糖的实验条件:熔融石英毛细管(50μm×34 cm);缓冲液为50 mmol/L硼砂/NaOH,pH值为10.0;电泳为10 kV,温度为60℃;检测波长为195 nm;进样为5000 Pa;时间3 s。此方法操作简便、准确度高、精密度好。该方法的测定结果与高效液相色谱法的结果较一致,样品中其他糖类对该方法不存在干扰,克服了莱因-埃农氏法测定乳糖不具专一性的缺点。
The disadvantage of present usual methods of determination of lactose is their limitations or inconveniences. In this paper, the optimal experimental conditions of determination of lactose in milk by high-performance capillary electrophoresis (HPCE, Agilent, HP3DG1601BA ) with UV detector were explored: fused silica capillary (50℃m ID×34 cm), buffer solution: 50 mM borate / NaOH, pH 10.0, HPCE conditions: 10 kV, 60 ℃, UV detection at 195 nm, sample was injected at 5 000 Pa for 3 s. This method is easy to operate and has good accuracy and precision. The result of this method is consistent with high-performance liquid chromatography (HPLC) method when they detect the lactose in milk, and other reducing sugars in milk will not interference the result of HPCE method.
