摘要
用拉曼光谱研究了γ射线诱变固体脱氧核糖核酸(DNA)结构的变化。拉曼光谱图显示磷酸二酯键伸缩振动谱带786cm(-1)、810cm(-1)和1100cm(-1)诱变后减弱,脱氧核糖或脱氧核糖与磷酸酯共同贡献的谱带884cm(-1)、1010cm(-1)和1068cm(-1)诱变后消失,脱氧核糖的1468cm(-1)峰诱变后显著增强,诱变后各碱基强度或频率发生了不同程度的变化,说明经过γ射线诱变以后,DNA碱基受损,主链断裂,双螺旋结构遭到破坏。
Structural changes of solid deoxyribonucleic acid (DNA) induced by gamma-ray are studied by Raman spectroscopy. By comparing the fore-and-aft Raman spectra plots of DNA induced by gamma-ray, the results are obtained: after DNA are induced by gamma-ray,the intensity of the Raman bands assigned to the flexing vibration of phosphodiester bonds at 786cm(-1)、810cm(-1) and 1100cm(-1) decrease, the Raman bands assigned to deoxyribose or deoxyribose and phosphodiester at 884cm(-1)、1010cm(-1) and 1068cm(-1) disappear, the deoxyribose marker line at 1468cm(-1) increases markedly, intensity and frequency of the marker bands of all four bases also happen changes differently degree. all of these indicate base damage, strand break, double helix structure encountering destroy.
