摘要
目的:研究高三尖杉酯碱和血栓通对培养人翼状胬肉成纤维细胞周期性变化的影响。方法:对翼状胬肉组织中的成纤维细胞进行培养并传代为第3代~5代。用高三尖杉酯碱(10-5mol/L)及血栓通(1.5g/L)分别处理24h,对照组用生理盐水。应用PIDNA荧光染色及流式细胞仪检测和分析细胞周期的变化。结果:高三尖杉酯碱组和血栓通组G1群体细胞明显增多,S期细胞明显减少。与对照组比较差异均有明显意义(P<0.01),其中高三尖杉酯碱更为明显。结论:高三尖杉酯碱和血栓通对翼状胬肉成纤维细胞的增生均有明显抑制作用,以高三尖杉酯碱更显著。其作用机理可能与抑制细胞DNA合成有关。
OBJECTIVE: To study the effect of homoharringtonine and xue shuan tong on cell
cycle of fibroblasts cultivated from human pterygium. METHODS: The fibroblasts derived from 3
to 5 generations of the cultured pterygium tissues. These cells had been treated with
homoharringtonine (10 -5 mol/L) and xue shuan tong1 5 g/L respectively, in the control group
the cells had been treated with normal saline. The changes of cell cycle were observe and
detected cells by the methods of propidium iodide (PI) staining and flow cytometry (FCM).
RESULTS:Compared with the control group, the distribution in cell cycle was showed that the
percentage of G1 cells was increased, and the S phase cells were decreased in the
homoharringtonine and xue shuan tong treated groups respectively. There was statistical
significant between the experimental group and control groups( P <0 01), and the effect of
homoharringtonine group was more obvious. CONCLUSIONS:The homoharringtonine and xue
shuan tong can significantly inhibit the proliferation of the fibroblasts derived from pterygium.
The mechanism of effect could be related with inhibition of synthesizing DNA.
