摘要
目的:建立同时测定北五味子中五味子醇甲、五味子醇乙、五味子酯甲、五味子甲素、五味子乙素含量的方法。方法:以Symmetry(C18色谱柱(4.6 mm×250 mm,5μm),流动相为甲醇-水,梯度洗脱,0~36 min(60∶40~66∶34);36~65min(66∶34~80∶20);65~70 min(80∶20),70~75 min(80∶20~100∶0),流速0.5 mL.min-1,检测波长254 nm。结果:五味子醇甲、五味子醇乙、五味子酯甲、五味子甲素和五味子乙素分别在0.021~4.160,0.020~4.000,0.021~4.240,0.020~3.960,0.021~4.200μg呈良好的线性关系(r=0.999 9);平均加样回收率分别为104.8%,104.2%,102.7%,104.6%,104.5%。RSD值均小于3%。结论:本方法稳定易行、重复性好,可用于北五味子药材的质量评价。
Objective: To develop a HPLC quantitative method for determination of schisandrin,schisandrol B,schisantherin A,deoxyschisandrin and γ-schisandrin in Fructus Schisandrae.Method: A symmetry C18 column(4.6 mm × 250 mm,5 μm) was used with methanol(A) and water(B) as mobile phases,in gradient elution.The gradient program was as follows: 0-36 min,changed from 60%A to 66%A,36-65 min,to 80%A,65-70 min,kept for 80%A,70-75 min,to 100%A.The flow rate was 0.5 mL·min-1 and detection wavelength was set at 254 nm.Result: The linearities of schisandrin,schisandrol B,schisantherin A,deoxyschisandrin and γ-schisandrin were in the ranges of 0.021-4.160(r=0.999 9),0.020-4.000(r=0.999 9),0.021-4.240(r=0.999 9),0.020-3.960(r=0.999 9) and 0.021-4.200(r=0.999 9).The average recoveries were 104.8%,104.2%,102.7%,104.6%,104.5%,respectively.Conclusion: The method developed in this study was reliable,and can be used for the quality control of the fruits of S.chinensis.
