摘要
目的观察4月龄和7月龄胎肝提取物(E4,E7)对BEL7402肝癌细胞增殖和分化的影响.方法采用显微镜下细胞计数,3HTdR掺入和MTT比色法,观察(10~1000)mg/L的E4和E7对培养中BEL7402肝癌细胞的影响.结果E4从100mg/L起可明显抑制7402细胞的生长和DNA合成,细胞生长抑制IC50为1444mg/L,DNA合成抑制IC50为2279mg/L.在(250~1000)mg/L时对整体细胞线粒体脱氢酶活力有明显的抑制作用,IC50为10130mg/L.E7在(10~100)mg/L对细胞生长和DNA合成具有明显的促进作用,至1000mg/L浓度未发现明显的抑制作用.在500mg/L和1000mg/L浓度时对整体线粒体脱氢酶活力具有一定的抑制作用.对细胞生长曲线影响,E4表现为500mg/L时能明显抑制7402细胞的生长,而在50mg/L时影响不明显;E7表现为500mg/L时影响不明显,而50mg/L时则有明显的促进作用.(100~1000)mg/L的E4能明显促进单个细胞脱氢酶的活力,而E7只在1000mg/L时有一定的促进作用,在(10~50)mg/L对额定细胞脱氢酶?
AIM To investigate the effect of the hepatic extracts (E 4,E 7) from 4 and 7 month old embryos on BEL 7402 hepatoma cell proliferation and differentiation in vitro . METHODS The effects of (10-1000) mg/L E 4 and E 7 on BEL 7402 hepatoma cells in culture were investigated by cell counting under microscope, 3 H TdR incorporation and MTT colorimetric assay. RESULTS E 4 obviously inhibited the proliferation and DNA synthesis of BEL 7402 hepatoma cells at (100-1000) mg/L concentrations, their half inhibitory concentrations (IC 50 ) were 144 4mg/L and 227 9mg/L respectively. E 4 also inhibited the total activities of mitochondrial dehydrogenase at (250-1000) mg/L ,the IC 50 was 1013 0mg/L . E 7 evidently promoted the proliferation and DNA synthesis of the cells at (10-100) mg/L ,and had no remarkable inhibitory effects until up to 1000mg/L , and also inhibited the total activities of dehydrogenase at (500-1000) mg/L . E 4 inhibited significantly the cell growth curve at 500mg/L and had no apparent effects at 50mg/L ;while E 7 had no remarkable effect on the cell growth curve at 500mg/L , and showed an obviously promoting effect at 50mg/L . E 4 at (100-1000) mg/L apparently promoted the dehydrogenase activities of a single cell. Only at 1000mg/L did E 7 have such promoting effects, at (10-50) mg/L E 7 had an obviously inhibitory effect on the mitochondrial dehydrogenase activity of definite number of hepatoma cells. CONCLUSION The embryonic cells express more differential factors to fit their differentiation at the early period, and cells produce more growth factors to fit their rapid proliferation at the late period.
