摘要
本文用MTT试验方法观察了4种体外传代培养的肿瘤细胞株(YAC—1、EL_4、MLA_(144)和BCMG细胞)对4种化疗药物(丝裂霉素-C、5-氟尿嘧啶、阿霉素和盐酸阿糖胞苷)的敏感性。结果表明:MTT代谢产物甲膨与细胞数量呈线性关系,此方法不仅可以反映不同化疗药物对同一肿瘤细胞的抑制特性不同,而且亦可测出不同的肿瘤细胞对同一化疗药物的敏感性存有差异。鉴于MTT法经济、直观、快速和简便,从而为进一步应用于临床非实体瘤的药敏检测奠定了方法学基础。
The MTT assay method was established and used for the rapid testing of cytotoxicity of different drugs(Mitomycin-C, 5-Fluorouracil, Adriamycin and cytos-sine arabinoside) against different non-solid tumor cell lines (YAC-1, EL-4, MLA144and bcmg cells). The result showed that there was a linear relationship between viable cell numbers and formazan production (expressed by optical density). This could show not only the suppressing patterns of one cell line to different chemotherapentic agents, but also different sensitivity of tumor cells to each drug. The results indicated that the MTT assay might offer a practical, economical, rapid, simple and very efficient method of assessing cytotoxicity of drugs in non-solid tumors.
