兔关节软骨细胞的体外分离、纯化及鉴定 被引量:9

摘要 体外培养软骨细胞是研究大骨节病、骨关节炎等软骨疾病的一个重要手段,也为软骨细胞移植修复软骨缺损、畸形提供了可能。本文通过一种简单易行的方法,获得大量软骨细胞,绘制了其生长曲线,体外培养的软骨细胞倍增时间为56.9小时,研究了软骨细胞的传代、纯化特点,通过GAG染色、Ⅱ型胶原染色对软骨细胞进行鉴定,用透射电镜对其超微结构进行了观察。 In vitro culture of chondrocyte is an important method to perform some research work on cartilage diseases,such as osteoarthrosis and osteoarthritis,and at the same time provide the feasibility to repair cartilage defect and malformation by chondrocyte transplantation. In this experiment we harvested a large number of chondrocytes through an easy way and studied biological behaviors of chondrocytes. The doubling time of the 3rd passage of cells was 56.9 hours. Chondrocytes could be purified by two method and identified with safra-nine- 0 staining and type II collagen immunohistochemistry reaction. We also observed their ultrastructure by electronic microscope.
