摘要
目的:研究芩连四物汤的化学物质基础及其抗凝血生物效应。方法:采用水煎醇沉,醇溶液减压回收乙醇,用适量水溶解后,上D101大孔树脂采用不同浓度的乙醇梯度洗脱,各不同洗脱部位采用硅胶柱层析的方法分离,所得单体通过UV,IR,NMR及MS等方法确定其化学结构;将所得成分配成水溶液(100μmol.L-1),采用体外法测定对血浆凝血酶时间(TT)的影响。结果:从芩连四物汤中分离并鉴定了5个黄酮类成分、1个生物碱、3个芳香酸、1个倍半萜类成分及2个其它类化合物。黄酮苷元、芳香酸、生物碱及倍半萜类化合物均能显著延长血浆凝血酶时间(TT)。结论:首次对芩连四物汤整方进行化学物质分离和结构分析研究,其中9个化合物,包括千层纸素A、汉黄芩素、6-甲氧基汉黄芩素、小檗碱、黄芩素、阿魏酸、香草酸、咖啡酸、地黄苦苷元显示了一定的体外抗凝血活性。
Objective: To investigate the chemical constituents and anticoagulant activity of Qin-Lian-Si-Wu decoction. Methods: Qin-Lian-Si-Wu decoction was extracted by hot-water, and then concentrated by decompression, and added ethanol till the concentration, of ethanol reached 80%. The upper solution was concentrated at 40 ℃, dissolved by distilled water, separated by gradient elution with different concentration ethanol from macroporius adsorptive resins D101, then obtained the different fraction. Its constituents were isolated by silica gel column chromatographic method, and their structures were identified through UV, IR, NMR and MS spectrum. These compounds were prepared into aqueous solution, and then their effects on the blood plasma thrombin time in vitro in rabbits were determined. Result: Five flavonoids, one alkaloid, three aromatic acids, one sesquiterpene and two other components were isolated and identified from Qin-Lian-Si-Wu decoction. The results of thrombin time test showed that flavonoids, aromatic acids, alkaloid, sesquiterpenes could prolong the blood plasma thrombin time in vitro in rabbits remarkably. Conclusion: 12 compounds were isolated and identified from Qin-Lian-Si-Wu decoction for the first time. Nine compounds, including oroxylin A, wogonin, 6-dimethoxy wogonin, berberine, baicalein, ferulic acid, vanillic acid, caffeic acid, rehmapicrogenin showed the anticoagulant activity at a certain dosage
