黄芪通过抑制NF-κB减轻大鼠移植肝脏缺血再灌注损伤的实验研究 被引量:5

Astragalus membranaceu reduces ischemia-reperfusion injury of rat liver graft by inhibiting NF-κB

摘要 目的:探讨黄芪(AM)对大鼠移植肝脏缺血再灌注损伤的保护作用及机制.方法:选用128只雄性健康SD大鼠,随机分成黄芪给药组(AM组)和生理盐水对照组(NS组),以二袖套法建立原位肝移植模型.术后1,2,4h检测血清丙氨酸转氨酶(ALT)值,ELISA检测肿瘤坏死因子-α(TNF-α)值,WesternBlot检测肝组织细胞间黏附分子-1(ICAM-1)的表达,免疫组化法及ELISA测定肝组织核因子-κB(NF-κB)的表达,术后3d取肝脏标本做HE染色检查,TUNEL法检测肝脏细胞凋亡.结果:在肝移植完成后AM组ALT分别为(9.4±0.4),(10.8±0.5),(14.3±1.1)μkat/L,较NS组的(12.4±0.6),(15.9±0.8),(18.7±1.1)μkat/L低(P<0.05);AM组TNF-α水平分别为(757±54),(977±60),(318±53)ng/L,较NS组(1258±132),(1831±165),(499±66)ng/L低(P<0.05);AM组的NF-κB,ICAM-1水平在各个时间点均低于NS组(P<0.05);AM组肝细胞凋亡程度低于NS组(P<0.05);AM组肝脏组织的病理改变较NS组肝脏组织的损伤轻.结论:黄芪能减少炎症介质释放,减轻移植后肝脏结构和功能上的破坏,提高肝脏耐受缺血再灌注损伤的能力. AIM: To investigate the protective role of astragalus mcmbranaceu( AM ) in ischemic repcffusion injury of rat liver graft and its mechanism. METHODS: One hundred and twentyeight male Sprague Dawley (SD) rats were randomly divided equally into AM group and NS group and were used as donors and recipients of orthotopic liver transplantation. One hour, 2 h and 4 h following liver transplantation, samples of blood and hepatic tissues were taken for assay. Amounts of TNF-α ( tumor necrosis factor-α) and ALT (alanine aminotransferase) values in serum were detected. The activity of NF-κB (nuclear factor kappa B ) was determined by immunohistochemical staining and ELISA, and the expression of ICAM-1 (intercellular adhesion molecule-1 ) protein level was determined by Western Blotting. Three days following the transplantation, apoptosis indexes were observed by TUNEL. RESULTS: In AM group ALT values were (9. 4 ± 0.4),(10. 8±0.5) and(14.3 ±1. 1) μkat/L respectively, which were lower than ( 12.4 ±0.6 ) , ( 15.9 ± 0.8 ) and ( 18.7 ± 1.1 ) μkat/L respectively in NS group(P 〈 0.05 ). TNF-α concentrations were ( 757 ± 54 ), ( 977 ± 60) and ( 318 ± 53 ) ng/L, which were also lower than( 1258 ± 132), ( 1831 ±165) and (499 +66) ng/L respectively in NS group(P 〈0.05). Expression of ICAM-1 and the activity of NF-κB in AM group were also lower than those in NS group (P 〈 0. 05 ). The apoptosis indexes in the AM group were lower than those in NS group ( P 〈 0.05 ). Histological examination revealed that the injuries of hepatic tissues in NS group were more severity than those in AM group. CONCLUSION: Astragalus membrarlaceu protects hepatic I/R injury after liver transplantation, improves liver graft function and reduces the release of inflammatory mediators.
