IκB激酶在肝脏缺血再灌注中的作用 被引量:1

摘要 目的:探讨IκB激梅(IKK)在肝脏缺血再灌注(HIR)中的作用. 方法:Wister大鼠随机分为对照组、缺血再灌注组(HIR 组)、PDTC保护组(HIR+PDT组).HIR组将肝左叶及肝中叶入肝血流阻断60 min后再灌注.保护组先经阴茎背静脉注射PDTC 120 mg/kg-1后立即依HIR组致伤,对照组仅显露肝中叶及肝左叶肝蒂,不阻断,自阴茎背静脉注射无菌生理盐水1 mL.分别采用原位杂交、EMSA、免疫组化及赖氏法检测再灌注后1、6、12 h IκB激酶表达、NF-κB活性、TNF-α的表达及血浆中ALT含量. 结果:损伤后IκB激酶表达水平、NFκB结合活性、TNF- α表达水平、血浆中ALT含量升高;HIR+PDTC组与HIR 组相比较,IκB激酶表达水平、NF-κB活性、TNF-α表达水平、血浆ALT水平降低. 结论:HIR后肝内IκB激酶可促进NF-κB的高水平激活引起肝内TNF-α等炎症相关细胞因子表达增强,从而导致肝脏损伤.干预IKK-NF-κB通路可能是防止HIR后急性肝脏损伤发生、发展的一个有效途径.
