摘要 测定细胞内Ca^(2+)浓度已成为研究脑水肿发生机制的焦点。本文采用Fura-2荧光标记法,测定冷冻伤性脑水肿时突触体内游离钙离子浓度变化,来探讨突触体的制备方法、Fura-2测定神经细胞突触体内Ca^(2+)浓度的原理、方法及Fura-2优缺点和其影响因素。 Measuring intracellular free calcium concentration ([Ca2+ ]1) is a key in the study of brain edema. The changes of the concentration of the free calcium within cortical neurons were investigated on the model of brain edema induced by freezing injury. [Ca2+]; was determined in calcium fluorescent indicator Fura-2/AM loaded neuronal synaptosomes by using mono-wavelength spectrofluorometer. The method for preparing synaptosomes and mechanism of measuring cytosolic free calcium concentration by fluorescent indicator Fura-2/AM were studied
