摘要
目的:观察鹿角方吸收成分对培养乳鼠心肌细胞胞浆游离钙的作用,探讨其强心机制。方法:采用血清药理学方法,使用钙荧光探针Fura-Z/AM观察鹿角方药物血清对乳鼠心肌细胞胞浆游离钙的影响。结果:鹿角方药物血清可升高培养乳鼠心肌细胞胞浆游离钙浓度,其作用依赖于细胞外钙存在。结论:鹿角方吸收成分可促进细胞外Ca^(2+)内流,升高心肌细胞胞浆游离钙浓度,这与其增强心肌收缩有关。
Objective: To observe the effects of absorbale components of Lujiaofang (ALC)on cytosolic free calcium [Ca2+ ]i concentration in cultured immature rat myocardial cells, and investigate its cardial enhancement mechanism.
Methods:ACL- containing rat serum was collected by application seropharmacological method. [Ca2+ ]i concentration was measured by using calcium fluorescence probe Fura - 2/AM.
Results: ACL - containing serum elevated the concentration of cytosolic [ Ca2+ ] i in immature rat myocardial cells in the condition of extrocellular Ca 2+ .
Conclusion:Stimulation of ACL on Ca2 + intromission and elevation of myosolic[Ca2 + ]i concentration of myocardial cells are relation to enhancement of cardial constriction.
