摘要
通过对预先选择的7种表面活性剂作为谷氨酸发酵促进剂效果的初步筛选,得知两性甜菜碱449和芹菜素硬脂酸酯对产酸率提高有较为明显的效果;进而从加入时间、加入剂量两个方面考察了筛选出的两种表面活性剂对谷氨酸棒状杆菌(Corynebacterium glutamicum)T-613菌体生长和谷氨酸发酵产酸的影响,考察了两种表面活性剂同时加入的协同效果,得到了最佳发酵工艺参数。研究结果表明,菌种对数生长期(第3 h)加入质量分数为10%的两性甜菜碱449型表面活性剂2.5 mL,发酵中后期(第12 h)加入质量分数为0.5%的芹菜素硬脂酸酯0.8 mL,在不影响菌种生长的前提下,促使谷氨酸产酸率由3.05%提高到3.76%,较原产酸率提高了23.28%,发酵周期由44 h下降为36 h,缩短了8 h。
Comparing the effect of 7 pre-selected surfactants on glutamic acid fermentation, the results indicated that Amphiprotic betaine 449(AB449) and Pelargidenon 1449 stearate (PGS) can rise the rate of the glutamic acid production (RG) obviously. Then the effect of these two surfactants on cell growth and glutamate acid production by Corynebacterium glutamicum T -613 was investigated in respect of addition time and addition dosage. The optimal technological parameters were obtained by the cooperation effect of adding both surfactants. Experimental results showed that adding 10% AB449 2. 5 mL in 3 h at the strain growth logarithmic phase and 0. 5% PGS 0. 8 mL in 12 h since the beginning of the fermentation, under the condition of strain growing normally, the product yield rose from 3.05% to 3. 76%, and the productive efficiency was raised by decreasing the fermentation period from 44 h to 36 h.
