摘要
以枯草芽孢杆菌TM903为供试菌株,采用摇瓶分批补料发酵的培养方式,考察了甜菜碱添加量对枯草芽孢杆菌合成利巴韦林及其合成途径中嘌呤核苷磷酸化酶(PNPase)活力的影响.结果表明,初始培养基添加0.7.g/L的甜菜碱,发酵36 h后每间隔3 h流加0.1 g/L甜菜碱,可显著提高PNPase的相对酶活,并且使利巴韦林的产量达4.21.g/L,比未添加前提高了47.2%.
The activity of PNPase and ribavirin production via Bacillus subtilis TM903 was investigated by fed-batch fermentation in shake flasks with the different concentration of betaine. 0.7 g/L betaine was added to the initial fermentation medium,and 0.1 g/L betaine was added into the fermentation system every three hours after 36 hours. The results show that the activity of PNPase can be improved markedly and ribavirin yield raise by 47.2% (4.21g/L) compared with the fermentation system without betaine.
