摘要
利用复合酶水解蚕蛹蛋白制备小分子多肽,通过正交实验得到水解工艺的优化组合为:木-Fla复合酶,酶用量为0.5%,底物含量为4%(1∶25),酶解pH为7.0,反应温度为60℃,酶解时间为3h,在此条件下水解,小分子多肽得率为32.16%,同时建立了毛细管凝胶电泳测定蚕蛹多肽分子的检测方法。碳粒廓清实验表明:蚕蛹蛋白酶水解液中小分子多肽可增强小鼠网状内皮系统吞噬功能。
The low molecular peptides were prepared from silkworm pupa protein by hydrolysis with compound enzyme. The results of orthogonal test showed that the optimum hydrolysis conditions was :the blend of proteolytic enzymes was papain and flavourzyme,the ratio of enzyme to substrate (E/S) was 0.5 % (w/w) ,the substrate concentration was 4,0 % (w/v), the hydrolyzing reaction occurs at 60℃and pH 7.0 for 3h. The yield of low molecular peptides was 32.16 % under the conditions. Carbon clearance test showed the low molecular peptides preparation from silkworm pupa protein could strengthen the ability of macrophage phagocytosis.
