大孔吸附树脂分离纯化草珊瑚总黄酮的工艺研究 被引量:8

摘要 目的研究大孔吸附树脂分离纯化草珊瑚总黄酮工艺,为草珊瑚总黄酮的工业化生产提供实验依据。方法以贵州产草珊瑚为原料,以草珊瑚总黄酮含量及回收率等为考察指标,选用大孔吸附树脂对草珊瑚总黄酮进行分离纯化,分别采用静态试验、动态试验等考察大孔树脂对草珊瑚总黄酮的分离纯化效果及影响因素:结果AB-8型大孔吸附树脂对草珊瑚总黄酮静态饱和吸附量为116.25mg·g^-1(干树脂),洗脱率92.9%,动态饱和吸附量为94.5mg·g^-1(干树脂),总黄酮回收率在96.1%、纯度在90%以上,是实验树脂中分离纯化草珊瑚总黄酮的最佳大孔吸附树脂。结论分离纯化草珊瑚总黄酮最佳工艺条件为:AB-8型大孔吸附树脂,洗脱剂为70%乙醇,洗脱剂用量为3倍树脂体积,流速3~4ml/min,上柱总黄酮量与树脂比为1:10.5,上柱液总黄酮浓度为19.8mg/ml,流速2~3ml/min,冲洗杂质用水体积2~3BV,上柱液pH值4~5。 Objective A method for separation and purification of the flavonoids from Sarcartdrae glabra with macroporous resin was studied. Methods By using Sarcandrae glabra in Guizhou and with the content and recovery rate of flavonoids as indexes and the static absorption capacity and the static elution ratio were observed. Results The results were as flollows : The static absorption capacity of AB -8 type resin was 116.25 mg·g^-1, the static elution ratio was 92.9% , the dynamic absorption capacity of AB -8 type resin was 94.5 mg·g^-1, the recovery rate was more than 96.1% and the purity was more than 90%. AB -8 type resin was the best for separating and purificating Sarcandrae glabra in flavonoids. Conclusion The optimum conditions is : AB - 8 type macroporous resin, the eluant is 70% alcohol and 3 times as the volume of the resin, the volume of the resin is 10.5 times of flavonoids in sample, concentration of flavonoids of sample is 19.8 mg·g^-1 and current velocity of 2 - 3 mg·g^-1, pH value of sample is 4 -5.
