强骨丹对去卵巢骨质疏松大鼠股骨作用的研究

摘要 目的:观察中药强骨丹对去卵巢骨质疏松大鼠股骨骨量的影响,探讨其防治绝经后骨质疏松症的作用。方法:选取Witar雌性大鼠50只,随机分为假手术组、模型组、强骨丹高、低剂量组及西药组5组,每组10只。摘除卵巢后第4天分别给予生理盐水、强骨丹高、低剂量和二膦酸盐,各组均在服药14周后处死动物,进行股骨骨密度测定和骨组织形态计量学观察。结果:模型组骨密度降低(P<0.01),骨小梁体积降低(P<0.01),骨吸收表面上升(P<0.01),成骨表面、类骨质表面、类骨质宽、四环素单双标记表面、矿化沉积率均明显上升(P<0.01),骨重建时间缩短(P<0.05)。强骨丹高剂量组骨密度较模型组增加(P<0.05),骨吸收表面降低(P<0.01),成骨表面亦降低(P<0.05);强骨丹高、低剂量组骨小梁体积增加(P<0.01),四环素单双标记表面显著减少(P<0.05或P<0.01),矿化沉积率降低(P<0.01),矿化时间和骨重建时间均延长(P<0.01)。结论:强骨丹能够防止去卵巢大鼠股骨下端骨组织的丢失,其作用是通过抑制骨吸收,降低过高骨转换率而实现的,对绝经后骨质疏松症有防治作用。 Objective: To observe the effect of Chinese medicine Qianggu Dan on femur of ovariecomized rats and study its anti-osteopototic mechanism. Methods: Ninety femal rats were randomly divided into 5 groups: sham operation group (Group 1), model group (Group 2), Qianggu Dan Group with high dosage (Group 3), Qianggu Dan Group with low dosage (Group 4) and western medicine group (Group 5). Each group has 18 rats. NS, high dosage Qianggu Dan, low dosage Qianggu Dan and biphosphnates were given to Group 2, 3, 4, 5 respectively. All rats were killed 14 days after treatment. Bone mineral densities ( BMD) and bone histomorphometry in femur were measured. Results: In Group 2, bone mineral density and bone trabecula volume were decreased (P<0.01), TBCS, TBOS, TOS, MOSW, STS/DTS and iMaR were increased markedly (P<0.01) and the period of bone rebuild was shortened (P<0.05). In Group 3, bone mineral density, BCS and TBOS were lowered (P<0.01or 0.05). In both Group 3 and 4, the following were improved: bone trabecula volume (P<0.01), STS/DTS (P<0.05 or 0.01), iMaR (P<0.01) and the period of bone rebuilding (P<0.01). Conclusion: Qianggu Dan can prevent the loss of the distal femur of ovariectomized rats through inhibiting bone absorption and decreasing the rate of bone turnover. Qianggu Dan can prevent and treat osteoporosis.
