摘要
观察促甲状腺素释放激素(TRH)在体内和体外对小鼠脾细胞产生IFN和IL-2的影响。结果表明,正常小鼠每天腹腔注射TRH0.2μg/只,共10d,其脾细胞受ConA刺激产生的IFN和IL-2产量无显著增高(P>0.05);对氢化可的松处理的免疫抑制小鼠,加用以上剂量TRH后,其脾细胞产生IFN和IL-2的能力明显提高(P<0.01)。在体外用TRH孵育小鼠脾细胞,其浓度在0.0039~1(10-10mol/L)时可显著提高ConA诱生的脾细胞IFN和IL-2的产量,且产量与TRH的浓度呈正相关;但高于此浓度时,其产量与TRH浓度呈负相关,呈现与剂量相关的双相调节作用。提示TRH的免疫调节机理之一是通过影响这些细胞因子的产生。
Studied the effect of thyrotropin releasing hormone(TRH) on the production of interferon(IFN) and interleukin 2(IL 2) induced by concanavalin A(ConA) in murine spleen cells in vivo and in vitro.The results showed that the products of IFN and IL 2 increased nonsignificantly(P>0 05),which induced by ConA in spleen cells of the normal mice treated I.P. with TRH of 0.2 μg a day for ten days,but the immunosuppressed mice after treatment with hydrocortisone were injected the same dose of TRH,in which the ability of spleen cells to product IFN and IL 2 rose notably(P<0 01).In vitro cultured the murine spleen cells with TRH and found that TRH at the concentration of 0.0039~1(10 -10 mol/L)significantly enhanced the products of IFN and IL 2,and there was a positive correlation between the products and TRH dosage,but TRH at the concentration beyond 1×10 -10 mol/L remarkably reduced the products of IFN and IL 2,and a negative dose effect relationship was obtained.It indicated that TRH has doublephase regulatory effect on the production of these cytokines in murine spleen cells.Thus,it suggest that one of the immunoregulative mechanisms of TRH seems to effect the production of cytokines.
