甲醛染毒小鼠睾丸DNA蛋白质损伤及相关基因蛋白质表达的变化 被引量:1

摘要 目的研究甲醛对小鼠睾丸细胞DNA损伤和交联作用及相关基因Bcl-2、Bax蛋白质表达的影响。方法用0.2~20.0mg/kg的甲醛对小鼠进行染毒,采用彗星实验检测小鼠的睾丸细胞DNA损伤及DNA蛋白质交联;进一步应用免疫组化技术结合显微镜图像分析技术检测睾丸细胞的Bcl-2和Bax的表达情况。结果低浓度(0.2mg/kg)的甲醛能引起小鼠睾丸细胞明显的DNA损伤,高浓度(20.0mg/kg)的甲醛引起DNA蛋白质交联。各浓度处理组Bax阳性表达率明显增多(P<0.05),而各浓度处理组Bcl-2的阳性表达率明显低于对照组(P<0.05)。结论甲醛能引起DNA损伤和交联,以及Bcl-2、Bax的表达发生显著性改变。 Objective To observe the damaging effect of formaldehyde on DNA and the expression related genes of Bcl-2 and Bax in mice testis. Methods Mice were exposed to formaldehyde at the doses of 0. 2 - 20.0 mg/kg. Comet test was used to investigate DNA damage and DNA protein crossiag in mice testis. Immunity technology and microscope image analysis technology was used to further explore testis' Bcl-2 and Bax expression. Results At the dose of 0. 2 mg/kg formaldehyde caused obvi- ous DNA damage in mice testis while 20 mg/kg formaldehyde could cause DNA protein crossing. Compared with the control, the positive expression rates of Bax in formaldehyde groups increased obviously, while the positive expression rates of Bcl-2 decreased(P〈0.05). Conclusion It was suggested that formaldehyde could induce DNA damage and DNA eressing. At the same time, the expression of Bax and Bcl-2 could present obvious expression changes.
