摘要
目的:通过观察雷公藤多苷对UUO小鼠肾小管间质细胞表型转化的影响,探讨抗风湿中药在防治肾间质纤维化中的作用。方法:采用小鼠单侧输尿管结扎(UUO)模型,用雷公藤多苷进行干预,用血管紧张素转化酶抑制剂蒙诺作为对照。肾组织中α-平滑肌肌动蛋白(α-smoothmuscleactin,α-SMA)、波形蛋白(Vimentin)蛋白表达采用免疫组化或Western-blotting方法检测;基因表达采用逆转录-聚合酶链式反应(RT-PCR)方法。结果:UUO模型组小鼠肾间质纤维化程度以及α-SMA、Vimentin蛋白表达和基因表达水平均较假手术组显著增高,而雷公藤和蒙诺治疗均可显著降低肾间质细胞表型转化。结论:抗风湿药雷公藤多苷可显著降低肌成纤维细胞的积聚,可能是防治肾间质纤维化的分子机制之一。
Objectlve: To investigate the effects of Tripterygium Wlilfordii on transdifferentiation of renal interstitial cells in mice kedney with the Unilateral Ureteral Obstruction(UUO) by examining the expression of α - smooth muscle actin(at-SMA), Vimentin. Methods: Mice were randomly assigned to shame operation group (shame proup ), UUO group, Tfipterygium Wlilfordii and ACEI treatment group after UUO. The protein expression of α- SMA, Vimentin were assessed by immunohistochemistry and Western - blotting;at - SMA,Vimentin mRNA were detected by using reverse transcription- polymerase chain reaction analyses( RT -PCR). And the change of renal pathology was observed at the same time. Results: In comparison with the shame group, the protein levels of at - SMA, Vimentin of UUO mice and treatment groups increased significantly, meanwhile Tripterygium Wlilfordii and ACEI could significantly reduce the expression of at - SMA, Vimentin on levels of protein. The effects of Tripterygium Wlilfordii on Vimentin were more significant than ACEI. Conclusion: Tripterygium Wlilfordii could reduce the transdifferentiation of renal interstitial cells and prevent renal interstitial fibrosis.
