摘要
目的建立一种流动注射荧光法测定人体血液中叶酸含量的新方法。方法在流动注射系统中,用自制的透析分离器在线分离血液样品,二氧化铅固相反应器在线氧化叶酸为强荧光物质,在激发波长281nm、发射波长450nm处测定氧化产物的荧光强度。结果测定叶酸的线性范围为5×10^-8-6×10^-6/mL,对浓度为3.00×10^-7g/mL的叶酸标准溶液平行测定11次的相对标准偏差为2.2%,透析分离器的透析率为23%,对3个不同人体血浆样中叶酸测定的回收率为95.5%-108.0%。结论该法简单、快速、灵敏、准确,能够满足血液中药物分析的要求。
Aim To develop a new flow injection fluorimetric method for the determination of folic acid in human blood. Methods Combining with dialysis separation sample, folic acid is on-line oxidized into a strong fluorescent compound with maximum excitation wavelength of 281 nm and emission wavelength of 450 nm with a lead dioxide solid-phase reactor, and the fluorescence intensity of oxidation product is proportional to the concentration of folic acid. Results Linear range for determination of folic acid is from 5 × 10^-8to 6 × 10^-6g/mL. The relative standard deviation is 2. 2% for 11 repetitive determinations of 3.00 × 10^ -7g/mL folic acid and the sample throughput is 25 h^ -1. The recoveries of folic acid in human blood are 95.5% - 108%. Conclusion The proposed method is sim- ple, rapid, sensitive and accurate, and suitable to be applied to the determination of folic acid in human blood.
