摘要
以KMnO4 为氧化剂 ,在pH 4 .2 ,80℃水浴条件下 ,以 2 89nm为激发波长 ,在 4 4 0nm处测定叶酸氧化产物喋呤 6 羧酸的荧光强度 ,从而间接测定叶酸的含量 ,叶酸的检测限为0 74 μg L ,较直接荧光法测定叶酸的灵敏度提高了 12 5倍。利用本法测定叶酸片剂的含量 ,回收率为 10 3 3%~ 114.0 %之间 ,相对标准偏差小于 1 8%。
The optimum conditions of the fluorimetric determination of pterin 6 carboxylic acid, oxidation product of folic acid using potassium permanganate as oxidant, were studied. The interference of coexistence matter was investigated. On this basis, the fluorimetric method for indirect determination of folic acid was established. The experimental result indicated that when the potassium permanganate solution was added to the pH 4.2 sample solution and heated on the 80℃ water bath for 4 minutes and the H 2O 2 was added to eliminate excessive permanganate, 289 nm and 440 nm were chosen as the excitation and emission wavelengths. The detection limit of folic acid was 0.74×10 -9 g/mL. The sensitivity was increased 12.5 times over the method of direct fluorimetric determination of folic acid. The method was used to determine the amount of folic acid in tablets. The recovery was 103.3%~114.0% with a relative standard deviation below 1.8%.
