断肢再植缺血再灌注损伤的保护机制 被引量:4

摘要 目的研究兔断肢再植离断肢体骨骼肌缺血再灌注损伤的保护机制。方法建立家兔断肢再植模型,随机分3组:灌流液灌注组、缺血再灌注组和对照组,灌流液灌注组先用灌注液对离断肢体进行灌注,然后进行再植;缺血再灌注组不进行灌注直接再植通血,对照组不做处理,两实验组分别于通血后5 h取材。测定血清一氧化氮(NO)、血浆内皮素-1(ET-1)含量及骨骼肌丙二醛(MDA)、超氧化物歧化酶(SOD)、线粒体ATP酶活性,观察骨骼肌损伤情况。结果灌流液灌注组ATP、MDA、SOD、NO和ET-1值等均较缺血再灌注组有明显改善。结论再植前对离断肢体进行预灌注,可以改善血管活性,减轻再植肢体后骨骼肌的缺血再灌注损伤程度。 Objective To study the protection mechanism of skeletal muscle' s ischemia repeffusion injury (IR) after limb replantation. Methods 30 rabbits were randomly divided into three groups: the perfusation group, IR group and control group. After establishing the amputated replantation models, the perfusation group was perfused with perfusion solution before replantation, the IR group was not perfused. Five hours later, the contents of malondialdehyde(MDA), superoxidedismutase(SOD), ATP enzyme of mitochondrial of skeletal muscle and content of the blood plasma ET-1 and blood serum NO (NO^2-/ NO^3-) were measured. Results In the perfusation group. The improvement was significant compared with the IR group. Conclusion The IR of skeletal muscle may be relieved and the activity of blood vessel could be improved if the amputated limbs are perfused with perfusion solution before replantation, relieving the IR injury.
