摘要
目的研究兔股动脉平滑肌组织中Ca2+-ATP酶、琥珀酸脱氢酶(SDH)活性和Ca2+、Mg2+含量在缺血期间的变化及其与缺血性血管痉挛的关系。方法在新西兰大白兔的断肢缺血动物模型中,使用组化技术检测股动脉平滑肌组织中Ca2+-ATP酶、SDH活性的变化,用图像分析系统测定其光密度来代表酶活性;用原子吸收光谱分析法测定股动脉平滑肌组织中Ca2+、Mg2+含量的变化。结果缺血8小时,Ca2+-ATP酶、SDH活性下降显著(P<0.05),缺血16小时后,Ca2+-ATP酶、SDH活性下降非常显著(P<0.01),缺血8小时后,Ca2+含量升高非常显著(P<0.01)。结论血管平滑肌组织中Ca2-ATP酶、SDH活性下降及Ca2+升高是缺血性血管痉挛的重要原因。
Objective To observe the changes of Ca2+ -ATPase and succinate dehydrogenase(SDH) activity and Ca2+ and Mg2+ contents in smooth muscle of femoral arteries in rabbit during ischemia,and to study the relationship between these changes and vasospasm. Methods On the ischemic model of theamputated limbs of New Zealands rabbits, the changs of Ca2+ - ATPase and SDH activity in smooth muscle offemoral arteries were detected with histochemistry method, the image pattern analysis system was used tomeasure optical density which stood for enzyme activity, the changes of Ca2+ and Mg2 +contents in smoothmuscle of femoral arteries were detected with atomic absorption spectroscopy analysis method. Results TheCa2+ -ATPase and SDH activity significantly decreased at 8 h after ischemia took place (P<0.05), and thedecrease became more significantly at 16 h after, ischemia happened (P<0.01). The content of Ca2+ sig-nificantly increased at 8 h after ischemia took place (P<0. 01). Conclusion The decrease of the Ca2+ -ATPase and SDH activity and the increase of Ca2+ content in vascular smooth muscle were important causeof ischemic vasospasm.
