北美黄连碱在体外试验中(浓度为10μg/ml、20μg/ml、40μg/ml)经过72h,原头节死亡率分别为59.1%66.4%、72.3%,较阿苯达唑、甲苯达唑、吡喹酮为高;扫描电镜观察到原头节多呈外翻型,顶突失去正常构型,头钩脱落,吸盘变形。北美黄连碱治疗小鼠继发性包虫病(3.75mg/kg/day×20,ig),囊肿抑制率达80.5%(P<0.001);扫描电镜观察到棘球蚴生发膜角质层面的微毛断裂脱落,有许多大小不等、深浅不一、边缘不整齐的陷窝,生发膜囊腔面的乳头状突起排列紊乱,出现许多不规则细胞碎片,未见内生性育囊。
When protoseoleces of Echinococcus granulosus maintained in 1% M-199 were exposed to d-Hydrastine (10μg/ml, 20μg/ml and 40μg/ml) for 72 h, the dead rates of protoseoleces were 59.1%, 66.4% and 72.75, respectively. When protoseoleces were exposed to albendazole, mebendazol and praziquantcl, however, much less effects of the drugs were observed. Disruption of external plasma membrane, hook detachment and sucker deformity of protoscolices were demonstrated by scanning electron microscopy. Mice infected with secondary cysts of Echinococcus granulosus were treated orally with d-Hydasine at a daily dose of 3.75mg / kg for 20 days, the mean cyst weight of each treated group was significantly lower than that of the control group with a cyst inhibition rate of 80.5%, Under scanning electron microscope, many pits arising from the outer and inner surfaces of the germinal membrane were observed.
