摘要
为研究甘油对冷冻干燥保存红细胞的作用机制,测定不同时期冷冻干燥保存-复水的红细胞回收率及超氧化物歧化酶(Superoxide d ismutase,SOD)、谷胱甘肽过氧化物酶(G lutathione peroxi-dase,GSH-Px)、过氧化氢酶(Catalase,CAT)的酶活性。结果显示,保存在25℃下,前90天内的SOD、GSH-Px与CAT的酶活性变化不大,酶活率保持在90%以上;随保存时间延长,180天后细胞回收率由初始的83.5%降低至48.5%,SOD、GSH-Px、及CAT的酶活率分别降低了25.6%、21.5%和18.4%。本实验发现保存过程是引起冻干红细胞SOD、GSH-Px、及CAT酶活性下降的主要原因。
In order to understand the protective mechanism of the glycerol on the preservation of freezedried human red blood cells at room temperature (25℃) ,the effects of storage time on activities of antioxidant enzymes and recovery of freeze dried red blood cells were studied using spectrophotometer colorimetry. The results demonstrated that the glycerol has remarkable effects on the cell viability. The maximum viability of red blood cells is 83.5% after pretreatment with 40% glycerol solution, which is significantly higher than that of control group (0% glycerol). After 180 days storage, the recovery of freeze dried red blood cells decreased from 83.5% to 48.5% , and superoxide dismutase (SOD) , the glutathione peroxidase (GSH-Px) and the catalase (CAT) activity decreased by 25.6% ,21.5% and 18.4%. Protective enzymes activity showed a similar decreasing pattern with the recovery of freeze dried red blood cells during the preservation processes.
