摘要 目的;建立双波长高效液相色谱同时测定人血清中维生素A、E(VitA、VitE)浓度的方法。方法:移取血清,加入乙腈乙醇混合溶液及正己烷,旋涡混合、离心,分离正己烷层氮气吹干,残渣用流动相溶解后进样。流动相为甲醇-水(92:8);流速1mL·min^-1;柱温30℃,紫外检测器在325nm和292nm检测VitA和VitE。结果:VitA保留时间为2.63min。浓度线性范围50~800μg·L^-1,r^2=0.9995,平均回收率为97.3%(RSD〈5%),最低捡出量0.4ng(S/N〉3);VitE保留时间为7.61min,浓度线性范围2~16mg·L^-1,r^2=0.9999,平均回收率为96.2%(RSD〈6%),最低检出量2豫(S/,N〉3)。40例健康成人血清VitA平均值为(389.0±138.0)μg·L^-1。VitE平均值为(10.2±2.5)mg·L^-1。结论:双波长HPIC检测血清中VitA和VitE浓度的方法灵敏度高,回收率和重现性良好,操作快速、简便。 OBJECTIVE To establish an HPLC assay for the determination of vitamin A(VitA) and vitamin E(VitE) in hu man serum and evaluate the nutritional value of the body' s of VitA and VitE. METHODS The serum was extracted with n-hexane after the protein in the serum was deposited with acetonitrile/alcohol. The layer of n-hexane was dried with nitrogen, and the residue was dissolved with mobile phase and determined by HPLC. VitA and VitE were separated on XTerra^TM RP18 5μm,3. 9mm×150 mm column,detected at 292 nm for VitE and 325 nm for VitA. Using methanol / water(92/8) as mobile phase when the column temperature was 30℃. RESULTS The linearity range of calibration curve of VitA was 50 - 800μg·L^-2 (r^2 = 0. 999 5). The average extracted recovery is 97. 3% ,RSD〈5%. The linearity range of calibration curve of VitE was 2 - 16 mg·L^-1 (r^2 = 0. 999 9). The average extracted recovery was 96. 2 %, RSD〈6%. The detection limit of VitA was 0. 4 ng and VitE is 2 ng (S/N〉3). The whole operation could be finished in less than 30 minutes. CONCLUSION The method is sensitive, rapid, accurate and simple.
