摘要
目的建立测定人血清中维生素A浓度的HPLC法。方法采用HypersilODS-C18柱为分析柱;甲醇∶水(95∶5,V/V)为流动相;流速为1.2mL·min^(-1);检测波长325nm。结果维生素A与人血清中其他成分分离良好,平均回收率为102.54%,日内、日间RSD均<10%;血清中维生素A浓度在20~800μg·L^(-1)范围内线性关系良好,r=0.9993。结论该法操作简便、结果可靠,适用于人血清中维生素A浓度的测定。
AIM To establish an assay method for vitamin A concentration in serum with HPLC. MOTHODS Hypersil ODS-C18 was packed in the analytical column; the mobile phase consisted of methanol-water (95:5, V/V); the flow rate was 1.2 mL·min^-1; determination wavelength was 325 nm. RESULTS Vitamin A was separated well from other ingredients in serum. The mean recovery of vitamin A was 102.54% and the relative standard deviation (RSD) within day and between day were all less than 10 %. The assay linearity was determined over the range of 20 - 800 μg·L^-1 in serum ( r = 0.999 3). CONCLUSION The method is found to be simple and accurate, the experiment will supply reference methods for the determination of concentration of vitamin A in serum.
