摘要
应用4株伪狂犬病毒(Pseudorabies virus,PrV)作免疫原,建立了23株分泌抗PrV的单克隆抗体(McAb)的杂交瘤细胞株。根据其免疫生物学特性可分为4个类型:(1)具有FA、ELISA和Western blot特性;(2)具有FA和ELISA特性;(3)具有FA和Western blot特性;(4)只有FA特性。根据ELISA和Western blot试验,McAb可将5株PrV分为两种不同的抗原群。应用此McAb建立并比较了3种检测PrV抗原的方法(FA、RPHA、ELISA)。FA和RPHA特异性强,与分离病毒的符合率分别为88.2%和76.5%。两者的阳性检测率统计学上无显著差异(P>0.05)。RPHA设备简单,操作简便,结果直观,更适于在基层单位推广应用。
Twenty three hybridoma cell liaes secreting McAbs against PrV have been established with 4 PrV strains as immunogens. On the basis of their immunobiological characters, the McAbs could be divided into 4 classes( 1 ) positive by FA, ELISA and Western blot, ( 2 ) positive by FA and ELISA; ( 3 ) positive by FA and Western blot; ( 4 ) positive by FA only. The results of experiments showed that five PrV strains could be classified into 2 different antigenic groups with the McAblO by ELISA and Western blot. Three methods, FA, reverse passive hemaggiutination(RPHA) and ELISA, have been used for detecting PrV antigen. And the results indicated that FA and RPHA are more sensitive;their concordance rates with virus isolation ( VI)were 88.2% and 76.5%, respectively. With the advantage of simplicity in performance, and no need of sophisticated equipment, RPHA is a suitable test for the diagnosis of pseudora bies in the field.
