摘要
检测小鹅瘟病毒(GPV)VP3基因真核表达质粒在小鼠中的免疫效果。大量提取本实验室已构建的含有GPV VP3基因的真核表达质粒pVAXⅠ/VP3和空载体pVAXⅠ,分两组进行两点肌肉注射免疫小鼠,共免疫4次。利用MTT法检测免疫小鼠的T细胞增殖活性,间接ELISA法检测免疫小鼠血清中GPV特异性抗体的水平。淋巴细胞增殖指数,免疫小鼠与正常小鼠差异不显著。间接ELISA检测结果表明,pVAXⅠ/VP3质粒免疫组小鼠血清中GPV特异性抗体水平,显著高于空载体对照组和阴性对照组。已构建的GPV VP3基因真核表达载体可以诱导小鼠产生明显的体液免疫,为小鹅瘟核酸疫苗的研究奠定基础。
To detect the immune effect of eukaryotic expression plasmid cloned with goose parvovirus(GPV) VP3 gene in mice. Eukaryotic expression vector with goose parvovirus(GPV) VP3 gene (pVAX I/VP3 ) constructed in our laboratory and empty vector plasmid pVAX I were extracted abundantly, two groups of BALB/c mice were injected by i. m. with these two plasmids at the two points respectively and immunized for four times. The T lymphocyte proliferation activity was detected by MTT,serum GPV specific antibody of the mice were detected by indirect ELISA. T lymphocyte proliferation test showed no significant difference between the indexes of immunized mice and the normal mice. The result of indirect ELISA showed that the serum GPV specific antibody level of mice immunized with pVAX I/VP3 plasmid improved significantly comparing with those of mice in empty vector control group and negative control group. That indicated that the constructed eukaryotic expression vector with GPV VP3 gene induced significant humoral immunity. The research made a foundation for developing goose plaque DNA vaccine.
