摘要
建立了测定茶水浸出物中咖啡碱含量的反相液相色谱法,并与分光光度法进行了比较,以探讨仪器色谱条件的选择及样品前处理过程。色谱实验采用Nova PakC18分析柱,以甲醇-水(2 5%HAc~2 0%NH4AC,体积比为90∶10)为流动相,在UV274nm处检测。样品采用直接进样(直接法)及除去杂质后再进样(间接法)的前处理方法,平均回收率为95 6%~102 6%,相对标准偏差小于4%(n=5),HPLC直接测定法较之文献法更准确、快速、简便。同时也探讨了HPLC法的应用前景。
An RP-HPLC method for determination of caffeine in water extracts contents of tea was introduced,compared with the spectrophotometric method.The choices of chromatographic experimental conditions and the sample processing program were discussed.The chromatographic experiments were carried out on a Nova-PakC_(18) column with methanol-water(2.5 %HAC-2.0 %NH_4AC,90∶10,v/v).The UV detection wavelength was fixed at 274nm.The tea samples were injected directly(the direct determination method)or injected after precipitation of the impurities(the indirect determination method)into HPLC system.The average recoveries of the additives were between 95.6 %-102.6 %,and the RSDs were lower than 4 %(n=5),The results showed the direct determination method using HPLC was more rapid,convenient and economical than the literature methods.
